Descripción
ANK3, also known as ankyrin-G, is a member of the ankyrin protein family that functions as a cytoskeletal adaptor protein, organizing and stabilizing membrane domains by linking integral membrane proteins to the spectrin-actin cytoskeleton. ANK3 is highly expressed in the nervous system, where it is critical for the proper localization and clustering of ion channels and adhesion molecules in neurons, particularly at the nodes of Ranvier and axon initial segments (AIS). This localization is essential for the initiation and propagation of action potentials, supporting neuronal excitability and communication. ANK3 also plays a role in synaptic stability and organization, contributing to proper brain development and function. Mutations in ANK3 are associated with neurological and psychiatric disorders, including bipolar disorder, schizophrenia, and autism spectrum disorders, where disruptions in neuronal signaling and ion channel organization occur. In cardiac muscle, ANK3 anchors critical ion channels and structural proteins, making it essential for maintaining cardiac excitability and rhythm. Dysregulation of ANK3 can lead to cardiac arrhythmias and neurodevelopmental abnormalities, highlighting its importance in both neuronal and cardiac cellular organization and function.
