Descripción
MC3R is a G protein-coupled receptor activated by melanocortins, including α-MSH, β-MSH, and ACTH. MC3R is expressed primarily in the hypothalamus, pituitary gland, and other peripheral tissues, where it plays a critical role in energy homeostasis, feeding behavior, and circadian rhythms. MC3R signaling involves coupling to Gs proteins, leading to increased cAMP levels and activation of downstream pathways that regulate appetite suppression and energy expenditure. In the hypothalamus, MC3R modulates the activity of neuropeptides, such as AgRP and POMC, balancing food intake and energy utilization. Dysregulation of MC3R has been linked to obesity, metabolic syndrome, and circadian rhythm disorders. MC3R also contributes to immune regulation, influencing anti-inflammatory responses and macrophage function. Emerging evidence highlights its role in skeletal muscle metabolism and cardiovascular function. Therapeutic targeting of MC3R is being explored for obesity treatment, metabolic disorders, and inflammatory conditions, emphasizing its importance in energy balance and homeostatic regulation.
