Background

SOCS1 is a critical negative regulator of cytokine signaling pathways, particularly those mediated by the Janus kinase/signal transducer and activator of transcription (JAK/STAT) system. It functions by directly binding to JAKs through its SH2 domain and targeting them for proteasomal degradation via its SOCS box, which recruits components of the ubiquitin-proteasome machinery. SOCS1 regulates immune responses by preventing excessive cytokine signaling, particularly in interferon-γ, IL-6, and IL-4 pathways, ensuring proper immune homeostasis and inflammation control.
