Keratin 8 (KRT8) is a type II intermediate filament protein primarily expressed in simple epithelial tissues, including the gastrointestinal tract, liver, and lungs, where it provides mechanical support and structural stability to epithelial cells KRT8 pairs with keratin 18 (KRT18) to form heterodimers that assemble into intermediate filaments, maintaining cytoskeletal integrity and cell resilience under mechanical and chemical stress KRT8 is involved in cell signaling, apoptosis, and protection against oxidative stress, contributing to epithelial cell survival Dysregulation or mutations in KRT8 are associated with liver diseases such as steatohepatitis, cirrhosis, and hepatocellular carcinoma, where KRT8 aggregates can impair cellular function KRT8 also participates in tumorigenesis by promoting epithelial-mesenchymal transition (EMT), enhancing cancer cell migration, invasion, and metastasis Elevated KRT8 expression is observed in various cancers, including colorectal, breast, and pancreatic cancer, where it serves as a diagnostic marker for disease progression and prognosis

This gene is a member of the type II keratin family clustered on the long arm of chromosome 12. Type I and type II keratins heteropolymerize to form intermediate-sized filaments in the cytoplasm of epithelial cells. The product of this gene typically dimerizes with keratin 18 to form an intermediate filament in simple single-layered epithelial cells. This protein plays a role in maintaining cellular structural integrity and also functions in signal transduction and cellular differentiation. Mutations in this gene cause cryptogenic cirrhosis. Alternatively spliced transcript variants have been found for this gene.

Keratins are a large family of proteins that form the intermediate filament cytoskeleton of epithelial cells, which are classified into two major sequence types. Type I keratins are a group of acidic intermediate filament proteins, including K9–K23, and the hair keratins Ha1–Ha8. Type II keratins are the basic or neutral courterparts to the acidic type I keratins, including K1–K8, and the hair keratins, Hb1–Hb6. KRT8 is often paired with keratin 18 in vivo.
