Descripción
TRAF4 is a unique member of the TRAF family that regulates signaling pathways involved in cellular processes such as survival, migration, and cytoskeletal organization. Unlike other TRAF proteins, TRAF4 interacts with various receptors, including TNFR superfamily members, Toll-like receptors, and growth factor receptors, linking them to downstream signaling cascades like Akt, MAPK, and NF-κB. TRAF4 is highly expressed during embryonic development and in epithelial cells, where it plays a role in tissue morphogenesis, cell polarity, and immune responses. It also participates in reactive oxygen species (ROS) regulation and ciliary function. Dysregulation of TRAF4 has been associated with cancer progression, where it promotes epithelial-mesenchymal transition (EMT), cell migration, and invasion. Knockout studies reveal defects in neural tube closure, abnormal epithelial development, and impaired ciliary function, emphasizing its roles in developmental processes and cytoskeletal regulation.
