Descripción
SH2D3A, also known as NSP1 (novel SH2-containing protein 1), is an adaptor protein containing an SH2 domain and a proline-rich region that mediates interactions with signaling molecules in receptor tyrosine kinase pathways. It is involved in regulating actin cytoskeletal remodeling, cell adhesion, and migration by linking signaling proteins like Crk-associated substrates (CAS) to downstream effectors. SH2D3A plays a key role in integrin-mediated signaling and growth factor receptor activation, particularly during cell motility and adhesion processes. It is highly expressed in tissues undergoing dynamic changes, such as embryonic development and wound healing. Dysregulation of SH2D3A has been linked to cancer progression, where it promotes metastasis and invasion through actin cytoskeletal reorganization and integrin activation. Knockout studies reveal defects in cell migration, adhesion, and spreading, underscoring its importance in cellular movement and signal transduction.
