Background
DENN domain containing 10 (DENND10) is a member of the DENN domain-containing protein family, which functions as guanine nucleotide exchange factors (GEFs) for small GTPases, particularly members of the Rab family. DENND10 is specifically involved in regulating intracellular vesicle trafficking by activating Rab proteins, facilitating membrane dynamics and cargo transport within the endosomal system. The DENN domain within DENND10 mediates interactions with GDP-bound Rab proteins, catalyzing their exchange to the active GTP-bound state. DENND10 has been implicated in processes such as endosomal recycling, cell signaling, and cellular homeostasis. Dysregulation of DENND10 has been linked to disorders involving disrupted vesicle trafficking, although its specific contributions to disease mechanisms remain an area of active research. Its broad expression pattern across tissues underscores its importance in maintaining intracellular transport and cellular function.
