Peroxisome proliferator activated receptor gamma (PPARG) is a nuclear receptor that plays a central role in the regulation of glucose and lipid metabolism, adipogenesis, and inflammation PPARG is highly expressed in adipose tissue, where it regulates the differentiation of preadipocytes into mature adipocytes and controls the storage of fatty acids PPARG also modulates the expression of genes involved in insulin sensitivity, glucose homeostasis, and lipid storage, and it is a critical factor in maintaining metabolic balance Dysregulation of PPARG has been associated with a variety of metabolic disorders, including type 2 diabetes, obesity, and cardiovascular disease PPARG is a major target for drugs like thiazolidinediones (TZDs), which are used to improve insulin sensitivity in type 2 diabetes However, PPARG activation also influences immune responses and inflammation, and its role in chronic inflammation has linked it to diseases such as atherosclerosis and cancer Modulating PPARG activity offers therapeutic potential for addressing metabolic diseases, inflammatory conditions, and cancer, making it an important target for drug development in multiple areas of medicine

PPARs form heterodimers with retinoid X receptors (RXRs) and these heterodimers regulate transcription of various genes. Three subtypes of PPARs are known: PPAR-alpha, PPAR-delta, and PPAR-gamma.
