PAX8 is a transcription factor required for the development of the thyroid gland, kidneys, and parts of the reproductive system. During embryogenesis, PAX8 regulates genes necessary for thyroid follicular cell differentiation and hormone synthesis, making it essential for thyroid gland formation and function. Mutations in PAX8 are associated with congenital hypothyroidism, thyroid dysgenesis, and impaired thyroglobulin production. PAX8 also plays a role in nephrogenesis, contributing to kidney development and epithelial cell differentiation. In cancers, PAX8 is frequently overexpressed in thyroid, ovarian, and renal cell carcinomas, where it serves as a diagnostic marker and regulator of oncogenic pathways. Knockout models show thyroid gland agenesis, defective renal development, and reproductive abnormalities, highlighting PAX8's role in tissue-specific gene regulation, organogenesis, and endocrine function.
