Nucleoporin 50 (NUP50) is another key nucleoporin involved in the transport of proteins and RNA across the nuclear envelope NUP50 functions as a shuttle protein in the process of nuclear import, facilitating the recycling of importin proteins, which are required for the delivery of cargo molecules into the nucleus It is involved in the release of cargo from importins at the nuclear pore and aids in the efficient transport of proteins involved in various cellular processes such as signal transduction, gene regulation, and DNA repair NUP50’s role in nuclear import is crucial for maintaining cellular function, as defects in this process can lead to impaired gene expression, cell cycle progression, and protein localization Dysregulation of NUP50 has been implicated in diseases such as cancer, where alterations in nuclear import and protein trafficking contribute to tumor progression and metastasis

NUP50 may serve as a binding site on the nuclear side of the pore complex for export receptor-cargo complexes. It interacts with multiple transport receptor proteins including p27Kip1. This interaction is required for correct intracellular transport and degradation of p27Kip1.
