Descripción

ALKBH1 is an alpha-ketoglutarate (α-KG)- and Fe(II)-dependent dioxygenase that functions in DNA and RNA repair, as well as histone demethylation. ALKBH1 removes methyl adducts, such as N3-methylcytosine (m3C) and N1-methyladenine (m1A), from nucleic acids, restoring their normal structure and preventing mutagenesis. Beyond its role in DNA and RNA repair, ALKBH1 acts as a histone H2A dioxygenase, regulating chromatin structure and gene expression through histone demethylation. It influences transcription, cellular differentiation, and embryonic development by modulating histone methylation marks. ALKBH1 has been implicated in cellular responses to oxidative and alkylating damage, as it counteracts methylation stress caused by environmental toxins or chemotherapeutic agents. Dysregulation of ALKBH1 is associated with cancer, where aberrant repair or histone demethylation may contribute to tumor progression, genomic instability, and drug resistance. ALKBH1 also plays a role in metabolic reprogramming by linking α-KG metabolism to epigenetic regulation.

Specifically demethylates DNA methylated on the 6th position of adenine(N(6)-methyladenosine) DNA. May play a role in placental trophoblast lineage differentiation(By similarity).
