Nuclear receptor subfamily 1 group D member 1 (NR1D1), also known as Rev-Erbα, is a nuclear receptor that plays a key role in regulating the circadian rhythm, metabolism, and inflammation NR1D1 functions by binding to DNA and regulating the expression of genes involved in lipid metabolism, glucose homeostasis, and inflammatory responses It works in opposition to other nuclear receptors, such as RORs (retinoic acid receptor-related orphan receptors), to control circadian clock genes and maintain the balance between the sleep-wake cycle and metabolic processes NR1D1 also has a role in regulating the immune system, where it modulates the expression of genes involved in immune responses and inflammatory pathways Dysregulation of NR1D1 expression or activity has been linked to metabolic disorders, such as obesity, diabetes, and cardiovascular diseases, as well as immune-related conditions like atherosclerosis and rheumatoid arthritis Its involvement in the regulation of daily metabolic rhythms also makes NR1D1 a potential target for therapies aimed at treating sleep disorders, metabolic diseases, and chronic inflammatory conditions
