Background

TET3 is a dioxygenase enzyme that converts 5-methylcytosine (5mC) to 5-hydroxymethylcytosine (5hmC), driving active DNA demethylation and playing a critical role in epigenetic reprogramming. TET3 is predominantly expressed during early embryonic development, oocyte maturation, and zygotic genome activation, where it facilitates global DNA demethylation and ensures proper gene activation for development. In somatic tissues, TET3 is involved in regulating neuronal differentiation, synaptic plasticity, and DNA repair pathways. TET3 plays a pivotal role in brain development and neurogenesis, where it ensures transcriptional flexibility required for cellular identity and maturation. Dysregulation of TET3 has been associated with neurological disorders, aging, and cancer, where altered DNA methylation patterns disrupt gene expression and cellular homeostasis. Knockout studies reveal defects in embryogenesis, impaired neurodevelopment, and increased genomic instability, underscoring TET3's essential role in DNA methylation reprogramming, neuronal function, and developmental regulation.
