LRRC14 is recognized as a regulator of transforming growth factor-beta (TGF-β) signaling and is predominantly expressed in immune and stromal cells where it modulates various biological responses linked to inflammation and fibrosis it functions as a TGF-β receptor-interacting protein acting as a negative regulator of TGF-β by preventing receptor activation and downstream Smad signaling pathways its expression is closely associated with cellular differentiation processes including the regulation of mesenchymal cell activity in wound healing and fibrotic tissue remodeling studies suggest that LRRC14 can influence tumor microenvironments by altering TGF-β activity contributing to either suppression or promotion of tumor growth depending on the context its dysregulation has been implicated in conditions such as cancer-related fibrosis and autoimmune diseases making it a target of interest for therapeutic modulation additional research is being conducted to better understand its interactions with other cytokine networks and its potential as a biomarker for fibrotic and oncologic disorders
