BAG cochaperone 6 (BAG6) is a protein that interacts with the HSP70 chaperone to regulate the turnover and degradation of misfolded proteins, playing a critical role in the cellular response to stress BAG6 is involved in maintaining cellular protein homeostasis, particularly in the degradation of unstable proteins through the ubiquitin-proteasome pathway It helps in protein quality control by preventing the accumulation of damaged or misfolded proteins, which can otherwise lead to cellular dysfunction BAG6 also regulates autophagy, a process through which cells degrade and recycle damaged proteins and organelles under stress conditions In cancer, BAG6 has been implicated in modulating apoptosis and promoting tumor cell survival by stabilizing key signaling proteins Dysregulation of BAG6 expression is linked to neurodegenerative diseases and heart disease, where defective protein turnover contributes to tissue damage and dysfunction BAG6’s involvement in stress responses, autophagy, and protein degradation highlights its importance in maintaining cellular health and stability

This gene was first characterized as part of a cluster of genes located within the human major histocompatibility complex class III region. This gene encodes a nuclear protein that is cleaved by caspase 3 and is implicated in the control of apoptosis. In addition, the protein forms a complex with E1A binding protein p300 and is required for the acetylation of p53 in response to DNA damage. Multiple transcript variants encoding different isoforms have been found for this gene.
