Descripción
KPNA6, also called importin-α7, is a nuclear transport adaptor protein that interacts with importin-β to mediate the nuclear import of cargo proteins containing classical nuclear localization signals (NLS). It plays key roles in cellular processes such as transcriptional regulation, cell cycle control, and DNA repair. KPNA6 is highly expressed in embryonic tissues, germ cells, and immune cells, where it supports cellular differentiation and developmental processes. It has been implicated in nuclear import during viral infections, where it facilitates the entry of viral proteins into the nucleus. Dysregulation of KPNA6 is associated with developmental defects, cancer progression, and immune dysfunction. Knockout studies reveal its critical function in early embryogenesis, nuclear transport efficiency, and maintaining proper gene expression regulation during cellular growth and differentiation.

The import of proteins containing a nuclear localization signal (NLS) requires the NLS import receptor, a heterodimer of importin alpha and beta subunits also known as karyopherins. Importin alpha binds the NLS-containing cargo in the cytoplasm and importin beta docks the complex at the cytoplasmic side of the nuclear pore complex. In the presence of nucleoside triphosphates and the small GTP binding protein Ran, the complex moves into the nuclear pore complex and the importin subunits dissociate. Importin alpha enters the nucleoplasm with its passenger protein and importin beta remains at the pore. The protein encoded by this gene is a member of the importin alpha family.
