Background

DENN domain containing 4C (DENND4C) is a guanine nucleotide exchange factor (GEF) that specifically activates Rab10, a small GTPase involved in vesicular trafficking, endosomal recycling, and membrane transport. DENND4C facilitates the GDP-to-GTP exchange on Rab10, enabling the active transport of cargo-containing vesicles to the plasma membrane or other intracellular compartments. It plays a significant role in regulating glucose transporter (GLUT4) recycling in adipocytes, which is essential for glucose uptake and cellular metabolism. DENND4C is also implicated in polarized transport in epithelial cells, ensuring proper protein and lipid distribution within specialized membrane domains. It is broadly expressed but shows elevated activity in metabolically active tissues such as muscle and adipose tissue. Dysregulation of DENND4C has been linked to metabolic disorders, including obesity and insulin resistance, where defective GLUT4 trafficking contributes to impaired glucose homeostasis. Its function in vesicle dynamics underscores its importance in cellular signaling, metabolism, and energy regulation.
