Descripción
DOCK2 is a hematopoietic-specific GEF expressed in leukocytes, activating Rac1/Rac2 to control chemotaxis and immune cell migration. It regulates lymphocyte development, T cell activation, and neutrophil responses to chemokines like CXCL12. DOCK2 interacts with ELMO1 to mediate actin polymerization, essential for leukocyte motility and immune responses. Dysregulation causes defects in lymphocyte trafficking and inflammation. In infections and cancer, DOCK2 can promote or suppress disease progression. Its activity depends on membrane lipids like PIP3 and phosphorylation. Knockout models show immunodeficiency and impaired immune surveillance, emphasizing DOCK2’s role in immunity.
