ANXA7 is a calcium- and GTP-dependent membrane-binding protein that plays a key role in exocytosis, membrane repair, and cell signaling. It acts as a calcium sensor, facilitating the fusion of vesicles with the plasma membrane, particularly in processes such as neurotransmitter release and hormone secretion. ANXA7 is expressed in the brain, heart, and other tissues, where it contributes to cellular calcium homeostasis and cell survival mechanisms. Mutations or dysregulation of ANXA7 have been associated with neurodegenerative diseases, such as Parkinson's disease, and cancers, where it can act as a tumor suppressor by regulating cell proliferation and apoptosis. Its roles in membrane dynamics and calcium-dependent signaling underscore its importance in cellular communication and homeostasis.

Annexin A7(Anx7) belongs to a ubiquitous and relatively abundant family of Ca2+-dependent membrane-binding proteins, which are thought to be involved in multiple aspects of cell biology including membrane trafficking, mediation of cell-matrix interactions and membrane organization within cells. Anx7, migrated as a 50 kDa protein in SDS-PAGE, has been proposed to function in the fusion of vesicles, acting as a Ca++ channel and as Ca++-activated GTPase, thus inducing Ca++ /GTP-dependent secretory events.

Annexin VII is a member of the annexin family of calcium-dependent phospholipid binding proteins.The Annexin VII gene contains 14 exons and spans approximately 34 kb of DNA. An alternatively spliced cassette exon results in two mRNA transcripts of 2.0 and 2.4 kb which are predicted to generate two protein isoforms differing in their N-terminal domain. ANXA7 is a protein with a molecular weight of approximately 51 kDa with a unique, highly hydrophobic N-terminal domain of 167 amino acids and a conserved C-terminal region of 299 amino acids. The latter domain is composed of alternating hydrophobic and hydrophilic segments. Structural analysis of the protein suggests that Annexin VII is a membrane binding protein with diverse properties, including voltage-sensitive calcium channel activity, ion selectivity and membrane fusion.
