Ragulator complex protein LAMTOR5 is a critical subunit of the Ragulator complex that anchors Rag GTPases to the lysosomal membrane, facilitating the activation of mTORC1 in response to amino acid and nutrient availability LAMTOR5 stabilizes the Ragulator complex structure and supports mTORC1 signaling, which is essential for regulating cellular metabolism, growth, and autophagy Its role in nutrient sensing and lysosomal positioning makes it vital for cellular homeostasis Studies highlight that loss or dysregulation of LAMTOR5 disrupts mTORC1 activation, leading to impaired protein synthesis, autophagy, and energy balance LAMTOR5 is also involved in immune responses, where mTORC1 signaling regulates T-cell activation and macrophage function Aberrant expression of LAMTOR5 has been linked to metabolic disorders, neurodegenerative diseases, and cancers, where impaired nutrient signaling promotes cellular dysfunction Recent studies suggest its involvement in stress response pathways and lysosomal biogenesis, reinforcing its essential role as a scaffold protein in coordinating lysosome-mTORC1 communication
