Descripción
GHRHR is a G protein-coupled receptor that binds growth hormone-releasing hormone (GHRH), regulating the synthesis and secretion of growth hormone (GH) from the anterior pituitary gland. GHRHR is primarily expressed in pituitary somatotroph cells, where its activation stimulates the cAMP/PKA and MAPK signaling pathways, leading to GH gene transcription and release. Growth hormone plays a critical role in growth, metabolism, and tissue repair by promoting the production of insulin-like growth factor-1 (IGF-1) in peripheral tissues. Mutations or dysregulation of GHRHR are associated with growth hormone deficiencies, leading to short stature, delayed growth, and metabolic imbalances. GHRHR has also been implicated in aging, where reduced GH signaling contributes to age-related declines in muscle mass and bone density. Therapeutically, GHRHR agonists are used to stimulate growth hormone production in patients with GH deficiency, while antagonists are being explored for treating acromegaly and certain cancers where GH overproduction contributes to tumor growth. GHRHR is a key target for modulating growth hormone signaling and managing growth and metabolic disorders.
