Background

GAB2 is a key adaptor protein that links receptor tyrosine kinases and cytokine receptors to intracellular signaling cascades, including PI3K/Akt, MAPK, and STAT pathways. It is predominantly expressed in hematopoietic cells, where it regulates immune cell activation, differentiation, and cytokine signaling. GAB2 interacts with GRB2, SHP2, and PI3K to modulate immune responses, hematopoiesis, and mast cell degranulation. Dysregulation of GAB2 is associated with inflammatory diseases, autoimmunity, and cancer, particularly leukemia, breast cancer, and melanoma, where it enhances cell proliferation, survival, and migration.
