Descripción
GRM2, or mGluR2, is a Group II metabotropic glutamate receptor primarily expressed in the central nervous system, including the hippocampus, prefrontal cortex, and thalamus. GRM2 couples to Gi/o proteins, inhibiting adenylate cyclase activity and reducing cyclic AMP (cAMP) levels, leading to decreased glutamate release and dampened excitatory signaling. GRM2 functions as an autoreceptor or heteroreceptor, modulating synaptic plasticity and neuronal excitability. It plays a significant role in cognitive function, emotional regulation, and stress responses. Dysregulation of GRM2 has been associated with psychiatric disorders such as anxiety, schizophrenia, and depression, where altered glutamate signaling disrupts synaptic balance. GRM2 agonists have shown promise as therapeutic agents for mood and anxiety disorders, as they reduce excessive glutamate release and normalize neuronal signaling. GRM2 is also being investigated for its role in addiction and neurodegeneration, making it a key target for psychiatric and neurological therapies.
