RAB10, member RAS oncogene family (RAB10) is a member of the small GTPase family, which plays an essential role in regulating intracellular vesicular trafficking and membrane dynamics RAB10 is particularly crucial in the regulation of vesicle transport from the Golgi to the plasma membrane, helping to control the proper trafficking of proteins and lipids across various cellular compartments It is involved in processes such as exocytosis, endocytosis, and the recycling of membrane proteins, thus ensuring the continuous supply of necessary components to the cell surface RAB10’s role in cellular functions extends beyond vesicular trafficking, as it also regulates the actin cytoskeleton and is essential for maintaining cell shape, polarity, and motility It is particularly significant in the regulation of cell migration and adhesion by controlling the movement of integrins, which are critical for the attachment of cells to their extracellular matrix RAB10 is also involved in the regulation of glucose uptake, especially in adipocytes and muscle cells, in response to insulin signaling, making it crucial for metabolic regulation and insulin sensitivity Disruption in RAB10’s function has been linked to diseases such as cancer, where abnormal cell migration and adhesion contribute to tumor invasion and metastasis, as well as type 2 diabetes, where impaired glucose uptake leads to metabolic dysfunction The significance of RAB10 in both metabolic and cellular signaling pathways underscores its importance as a potential therapeutic target for metabolic disorders and cancers

The small GTPases Rab are key regulators of intracellular membrane trafficking, from the formation of transport vesicles to their fusion with membranes. Rabs cycle between an inactive GDP-bound form and an active GTP-bound form that is able to recruit to membranes different set of downstream effectors directly responsible for vesicle formation, movement, tethering and fusion(By similarity). In parallel, it regulates the transport of TLR4, a toll-like receptor to the plasma membrane and therefore may be important for innate immune response. Plays also a specific role in asymmetric protein transport to the plasma membrane within the polarized neuron and epithelial cells. In neurons, it is involved in axonogenesis through regulation of vesicular membrane trafficking toward the axonal plasma membrane while in epithelial cells, it regulates transport from the Golgi to the basolateral membrane. Moreover, may play a role in the basolateral recycling pathway and in phagosome maturation.
