Dmx like 2 (DMXL2), also known as rabconnectin-3α, is a WD40-repeat protein involved in vesicle trafficking and signaling, with critical roles in neurotransmitter release, autophagy, and lysosomal function. DMXL2 functions as a scaffold protein, mediating interactions between Rab GTPases and their effectors, particularly in vesicle docking and fusion. It is heavily involved in the regulation of vesicular trafficking pathways in neurons and endocrine cells, where it modulates the release of neurotransmitters and hormones. DMXL2 is also linked to the regulation of the vacuolar-type H+-ATPase (v-ATPase), which is essential for lysosomal acidification and autophagic degradation processes. Its expression is highest in neural, endocrine, and metabolic tissues, reflecting its involvement in processes like synaptic signaling and energy homeostasis. Dysregulation of DMXL2 has been associated with neurodevelopmental disorders, diabetes, and lysosomal storage diseases, where defects in vesicular and lysosomal trafficking contribute to disease pathology. Its diverse roles in vesicle-mediated transport make DMXL2 a key regulator of intracellular dynamics and signaling pathways.
