Description

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptor is required for generation and long-term maintenance of T cell immunity. It binds to ligand CD70, and plays a key role in regulating B-cell activation and immunoglobulin synthesis. This receptor transduces signals that lead to the activation of NF-kappaB and MAPK8/JNK. Adaptor proteins TRAF2 and TRAF5 have been shown to mediate the signaling process of this receptor. CD27-binding protein (SIVA), a proapoptotic protein, can bind to this receptor and is thought to play an important role in the apoptosis induced by this receptor.

CD27, also known as TNFRSF7 (tumor necrosis factor receptor superfamily member 7), is a cell surface receptor primarily expressed on T cells, B cells, and NK cells, playing a pivotal role in the regulation of immune responses. CD27 is a member of the TNF receptor superfamily and functions as a key co-stimulatory molecule, contributing to the development and survival of T lymphocytes, the formation of memory T cells, and the activation of B cells. Its ligand, CD70, also plays an essential role in immune activation, particularly in T-cell-B-cell communication. CD27 engagement through CD70 induces signaling pathways critical for T-cell activation, proliferation, differentiation, and long-term immune memory. The CD27-CD70 axis is tightly regulated because dysregulation of these pathways can lead to immune system-related diseases, including autoimmune disorders, chronic inflammation, and cancer. In addition to its role in adaptive immunity, CD27 is involved in immune surveillance and the elimination of pathogens and tumor cells. Importantly, CD27 is a target for therapeutic intervention, particularly in immunotherapy strategies aimed at boosting the immune response against cancer.
