Calcitonin Receptor-Like Receptor (CALCRL) is a member of the G-protein-coupled receptor (GPCR) family and plays a critical role in diverse physiological processes by interacting with Receptor Activity Modifying Proteins (RAMPs). Its function depends on the specific RAMP partner, leading to the formation of multiple receptor subtypes with distinct biological roles. When CALCRL associates with RAMP1, it forms the Calcitonin Gene-Related Peptide (CGRP) receptor, which regulates vasodilation, pain transmission, and neurogenic inflammation, especially in migraine pathophysiology. Conversely, when paired with RAMP2 or RAMP3, CALCRL becomes a receptor for adrenomedullin (AM) and adrenomedullin 2/intermedin, respectively. These signaling pathways are vital for maintaining vascular tone, endothelial barrier integrity, and angiogenesis, especially during conditions like sepsis, heart failure, and ischemic injuries.
