Bcl-2-related ovarian killer protein (BOK) is a pro-apoptotic member of the Bcl-2 family that plays a critical role in mediating apoptosis through mitochondrial pathways BOK induces cell death by interacting with other Bcl-2 family members, promoting mitochondrial outer membrane permeabilization and facilitating the release of apoptotic factors like cytochrome c, which activate caspases and lead to programmed cell death Unlike other Bcl-2 family members, BOK is not regulated by the BH3 domain and has a unique role in apoptosis, particularly in ovarian cells BOK’s expression is tightly controlled in healthy tissues, and its dysregulation has been associated with various cancers, where it can influence tumor cell survival, apoptosis resistance, and cancer progression Additionally, BOK has been implicated in regulating neuronal cell death, and its role in maintaining cellular homeostasis under stress conditions suggests it may be a target for therapeutic strategies aimed at inducing apoptosis in tumor cells or protecting against neurodegeneration
