Thyroid hormone receptor alpha (THRA) is a nuclear receptor that mediates the effects of thyroid hormones (T3 and T4) by regulating gene expression THRA exists in multiple isoforms, which are generated through alternative splicing and have tissue-specific functions The receptor plays a crucial role in the development, growth, and metabolism of various tissues, including the brain, heart, and muscles THRA regulates cellular processes such as metabolism, thermogenesis, and lipid metabolism by influencing the transcription of target genes involved in these pathways The receptor also has a vital role in the development of the nervous system, particularly during fetal and postnatal development Mutations in THRA can lead to various thyroid-related disorders, including resistance to thyroid hormone (RTH), where tissues become less responsive to thyroid hormones, resulting in elevated thyroid hormone levels and metabolic disturbances Additionally, THRA dysregulation has been implicated in certain cancers, where altered thyroid hormone signaling can contribute to tumor growth and progression
