Description

Adapter protein which acts downstream of several membrane receptors including cytokine, antigen, hormone, cell matrix and growth factor receptors to regulate multiple signaling pathways. Regulates osteoclast differentiation mediating the TNFRSF11A/RANK signaling. In allergic response, it plays a role in mast cells activation and degranulation through PI-3-kinase regulation. Also involved in the regulation of cell proliferation and hematopoiesis.

Descripción

GAB2 is a key adaptor protein that links receptor tyrosine kinases and cytokine receptors to intracellular signaling cascades, including PI3K/Akt, MAPK, and STAT pathways. It is predominantly expressed in hematopoietic cells, where it regulates immune cell activation, differentiation, and cytokine signaling. GAB2 interacts with GRB2, SHP2, and PI3K to modulate immune responses, hematopoiesis, and mast cell degranulation. Dysregulation of GAB2 is associated with inflammatory diseases, autoimmunity, and cancer, particularly leukemia, breast cancer, and melanoma, where it enhances cell proliferation, survival, and migration. Knockout studies highlight defects in immune cell activation, mast cell responses, and hematopoietic development, demonstrating its importance in immune regulation.
