Description
Catalyzes the cofactor-independent reversible oxidation of gamma-hydroxybutyrate(GHB) to succinic semialdehyde(SSA) coupled to reduction of 2-ketoglutarate(2-KG) to D-2-hydroxyglutarate(D-2-HG). D,L-3-hydroxyisobutyrate and L-3-hydroxybutyrate(L-3-OHB) are also substrates for HOT with 10-fold lower activities.

ADHFE1 is a unique alcohol dehydrogenase that utilizes iron as a cofactor, distinguishing it from other zinc-dependent ADH family members. This enzyme is expressed in the brain and liver, where it plays an important role in metabolizing hydroxyacid substrates such as 4-hydroxybutyrate, also known as gamma-hydroxybutyric acid (GHB). ADHFE1 is particularly involved in pathways related to the metabolism of hydroxy acids, contributing to energy production and cellular detoxification.
