ADAM10, also known as ADAM metallopeptidase domain 10, is a type I transmembrane glycoprotein belonging to the ADAM (a disintegrin and metalloproteinase) family of proteins. It is encoded by the ADAM10 gene located on human chromosome 15. ADAM10 plays crucial roles in various biological processes, including cell adhesion, cell fusion, cell signaling, proteolysis, and ectodomain shedding of membrane proteins. It is involved in the cleavage of a wide range of cell surface proteins, including cytokines, growth factors, adhesion molecules, and receptors. ( Notch receptors, amyloid precursor protein (APP), cadherins, and several cytokines and growth factors). The proteolytic activity of ADAM10 is tightly regulated, as its dysregulation can have significant implications in various diseases, including cancer, Alzheimer's disease, inflammation, and cardiovascular diseases. ADAM10-mediated cleavage of Notch receptors is crucial for various developmental processes, and dysregulation of this process has been implicated in cancer and neurodegenerative diseases. ADAM10 has gained significant attention as a potential therapeutic target for various diseases due to its involvement in critical cellular processes and its ability to modulate the activity of numerous substrates. Inhibition or modulation of ADAM10 activity is being explored as a strategy for the treatment of diseases such as cancer, Alzheimer's disease, and inflammatory disorders
