Background

ACSL4 is an acyl-CoA synthetase with a strong preference for polyunsaturated fatty acids, such as arachidonic and docosahexaenoic acids, converting them into acyl-CoA derivatives for metabolic use. It is highly expressed in the brain, testis, and adrenal glands, where it supports processes requiring specialized lipid metabolism, including the biosynthesis of eicosanoids and membrane lipids. ACSL4 is critical in ferroptosis, a type of programmed cell death triggered by lipid peroxidation, through its role in incorporating polyunsaturated fatty acids into phospholipids. Dysregulation of ACSL4 has been linked to cancer, neurodegenerative diseases, and inflammatory disorders, as it influences processes such as oxidative stress and cellular survival.
