Background

ABL1 is a non-receptor tyrosine kinase involved in various cellular processes, including cytoskeletal remodeling, cell migration, DNA damage response, and signal transduction. It is ubiquitously expressed and localized in both the nucleus and cytoplasm, where it interacts with numerous signaling proteins. ABL1 contains SH2 and SH3 domains, a kinase domain, and regulatory regions that facilitate its interaction with other proteins and its activation in response to cellular stimuli. Dysregulation of ABL1, particularly through chromosomal translocations such as the Philadelphia chromosome, results in the BCR-ABL1 fusion protein, which drives chronic myeloid leukemia (CML) by promoting uncontrolled cell proliferation and survival. Targeting ABL1 with tyrosine kinase inhibitors, such as imatinib, has revolutionized the treatment of CML and highlighted its role as a key therapeutic target.
