Background

The ITGB1 protein consists of 798 amino acids and is a single-pass transmembrane glycoprotein that forms part of the integrin receptor complex. The structure of ITGB1 can be divided into three key domains:

Extracellular Domain:

  • The extracellular domain of ITGB1 spans approximately 700 amino acids and is responsible for mediating ligand binding when paired with an α subunit. This domain contains several cysteine-rich repeats and calcium-binding motifs, which are crucial for integrin activation and conformational changes required for ligand engagement.
  • This domain undergoes major conformational shifts between a bent, inactive state and an extended, active state. When integrins are in the bent state, ligand binding is prevented, but upon cellular signals or external stimuli, ITGB1 extends to interact with ECM ligands, facilitating cell adhesion and downstream signaling.
  • Ligands that bind to ITGB1-containing integrins include fibronectin, laminin, collagen, and vitronectin, highlighting its role in interacting with diverse matrix proteins.

Transmembrane Domain:

  • The transmembrane domain consists of a single hydrophobic α-helix of 20-25 amino acids that anchors ITGB1 into the plasma membrane. This domain plays a key role in transmitting signals from the extracellular matrix to the intracellular signaling machinery. The transmembrane helices of ITGB1 interact with the α subunit to maintain the integrin in its inactive state and undergo structural rearrangements upon activation.

Cytoplasmic Domain:

  • The cytoplasmic tail of ITGB1 is relatively short (~47 amino acids) but is highly significant for intracellular signaling. It interacts with various cytoskeletal proteins and signaling molecules, such as talin, kindlin, and paxillin, that link integrins to the actin cytoskeleton.   • ITGB1 integrins anchor cells to the ECM, maintaining tissue architecture and integrity. In epithelial tissues, ITGB1 is necessary for keeping cells attached to the basement membrane, thereby ensuring proper tissue organization and stability.

Cell Migration:

  • ITGB1 is a key player in cell migration, particularly in processes such as wound healing, immune cell trafficking, and cancer metastasis.
