Background

CD48, also known as SLAMF2 (Signaling Lymphocytic Activation Molecule Family Member 2), is a glycosylphosphatidylinositol (GPI)-anchored membrane protein that plays a crucial role in immune cell interactions and signaling. Protein Structure

  • GPI-Anchor: Unlike many other membrane proteins, CD48 lacks a transmembrane and cytoplasmic domain. Instead, it is anchored to the cell membrane via a GPI linkage at its C-terminus. This GPI-anchor facilitates the lateral mobility of CD48 within the lipid bilayer, enhancing its ability to participate in immune synapse formation.
  • Glycosylation Sites: CD48 has multiple N-linked glycosylation sites, which contribute to its stability, proper folding, and function in immune recognition.


Classification and Subtypes

CD48 belongs to the SLAM (Signaling Lymphocytic Activation Molecule) family, a subgroup of the immunoglobulin superfamily. Other members of this family include:

  • CD48 acts as a co-stimulatory molecule for T and B cells. Engagement of CD48 with its ligand 2B4 (CD244) on NK cells and other immune cells enhances their activation, proliferation, and cytokine production.
  • It plays a role in the formation of the immunological synapse, a specialized junction between T cells and antigen-presenting cells (APCs), facilitating efficient antigen recognition and immune response initiation.

Cytotoxicity and NK Cell Function:

  • CD48 is critical for NK cell-mediated cytotoxicity. Interaction between CD48 on target cells and 2B4 on NK cells promotes NK cell activation and the killing of infected or malignant cells.
  • This interaction is crucial in the immune surveillance against tumors and viral infections.

Regulation of Immune Responses:
