Apolipoprotein B (APOB) is a protein encoded by the APOB gene located on chromosome 2 in humans. It is one of the primary structural proteins of lipoproteins, specifically low-density lipoprotein (LDL) particles. The APOB gene undergoes a process called mRNA editing, resulting in two isoforms: APOB-100 and APOB-48. APOB-100 is synthesized in the liver and is a major component of LDL particles, which transport cholesterol and other lipids through the bloodstream. It plays a critical role in lipid metabolism by mediating the uptake of cholesterol by cells throughout the body, including peripheral tissues and the liver. APOB-100 acts as a ligand for the LDL receptor, facilitating the clearance of LDL particles from the circulation. APOB-48 is produced in the intestine and is essential for the assembly and secretion of chylomicrons, which are lipoprotein particles that transport dietary triglycerides and other lipids from the intestine to peripheral tissues. APOB-48 is generated through mRNA editing of APOB-100, resulting in a truncated form of the protein that lacks the LDL receptor-binding domain. The expression of the APOB gene is tightly regulated by various factors, including dietary nutrients, hormones, and transcriptional regulators. Factors such as sterol regulatory element-binding proteins (SREBPs), peroxisome proliferator-activated receptors (PPARs), and liver X receptors (LXRs) play key roles in modulating APOB gene transcription and lipid metabolism. Genetic variations in the APOB gene can impact plasma lipid levels and contribute to the development of cardiovascular diseases, such as atherosclerosis and coronary artery disease. Mutations that affect APOB protein structure or function can lead to dyslipidemia, characterized by elevated levels of LDL cholesterol and an increased risk of cardiovascular events.
APOB48 belongs to the apolipoprotein family, specifically derived from the APOB gene. It is a unique isoform generated through RNA editing by the enzyme APOBEC-1, which converts a cytidine to uridine at position 6666 of the APOB mRNA, creating a stop codon.
